Abstract

The behavior of not washing hands can increase the risk of suffering from diarrhea caused by microorganisms. Among these microorganisms are Staphylococcus aureus and Escherichia coli. It is necessary to innovate hand sanitizer from natural ingredients that are safer and do not irritate the skin. Single extracts of Miana leaves and Kemuning leaves have been scientifically proven to be efficacious as antiseptics due to the presence of flavonoid compounds, tannins, and saponins. This study aims to determine the best antiseptic combination of Miana leaves (Coleus scutellariodes (L) Benth) and Kemuning leaves (Murayya paniculata (L.) Jack) against Staphylococcus aureus and Escherichia coli in the form of hand sanitizer gel. The combined extract of miana leaves and kemuning leaves was made in three concentration variations, namely 5%:15%; 10%:10%; and 15%:5%. Then tested its activity as an antiseptic against Staphylococcus aureus and Eschericia coli. The combination that shows the optimum inhibition, is made of pharmaceutical preparations by optimizing the gel base in three variations of concentration (0.5%), (0.75%), (1%). The results showed that the optimum combination of Miana and Kemuning leaves was 15%:5% with an inhibitory power of 6-10 mm, and the best gel base concentration was 0.5%. The hand sanitizer gel formulation with the combination of miana and kemuning leaves was able to inhibit the growth of Staphylococcus aureus (7.3 mm) and Eschericia coli (9 mm) bacteria with the "moderate" category of inhibition. The hand sanitizer gel formulation of the combination of miana and kemuning leaves met the requirements for organoleptic tests, homogeneity, pH, dispersion, adhesion, drying time and viscosity for 28 days at room temperature.
