Abstract

Indonesia is a country that has a wealth of flora and fauna species. In Indonesia, there are about 30,000 types of plants and 7,000 of them have medicinal properties, namely dadap serep (Erythrina subumbrans) leaves. Dadap serep leaves are known to contain secondary metabolites in the form of flavonoids, saponins, and tannins. The content of secondary metabolites in dadap serep leaves can be used as anti-inflammatory, antipyretic, and antimicrobial. The purpose of this study was to determine the antibacterial activity of dadap serep leaf extract against Staphylococcus aureus bacteria and to determine the minimum zone of inhibition that could inhibit the growth of dadap serep leaf extract. This research was conducted with an experimental sample of dadap serep leaves extracted using the maceration method using 96% ethanol as a solvent. The positive control used was chloramphenicol while the negative control used was 96% ethanol, the active extract was tested to determine the Minimum Inhibitory Concentration (MIC) using visual observation and UV-Vis spectrophotometer.The results of the antibacterial activity test of Dadap serep leaf extract showed antibacterial activity against S. aureus ATCC 25923. The results of the inhibition zones at concentrations of 5%, 10% and 15% respectively were 15.83±1.040 mm, 17.66±0.288 and 18 ,50±1,732 mm. Dadap serep leaf extract which has the most active inhibition zone is at a concentration of 5%. Antibacterial activity is thought to come from the activity of flavonoid compounds, tannins and saponins in dadap serep leaf extract. The value of the minimum inhibitory concentration of dadap serep extract was 0.312%. Keywords: Erythrina Subumbrans, Staphylococcus Aureus, Minimum Inhibitory Concentration
