Abstract

In Indonesia, many plants have the potential to be used as traditional medicinal ingredients, one of which is the tamarind plant (Samanea saman (jacq.) Merr). The leaves of the trembesi plant have potential as antibacterial against Escherichia coli ATCC 25922 which is a pathogenic bacterium that causes infection in humans. One option to overcome an infection is to use antibiotics, because Escherichia coli is easily resistant to antibiotics and there are many negative impacts from long-term use of antibiotics, it is necessary to develop antibacterial agents by utilizing antibacterial active ingredients from medicinal plants. The purpose of this study was to determine the activity of the 70% ethanol extract of trembesi leaves (Samanea saman (jacq.) Merr) as an antibacterial of Escherichia coli ATCC 25922. The research method used is experimental. Trembesi leaf samples were extracted using maceration method with 70% ethanol, followed by fractionation test of n- hexane, aquadestilata, and ethyl acetate with a concentration of 10%. The positive control used was 250 mg chloramphenicol capsule and the negative control was each solvent fraction. Statistical analysis was performed using Shapiro-Wilk and One way Anova. The results of the antibacterial activity test of the trembesi leaf fraction showed that the trembesi leaf fraction had antibacterial activity against Escherichia coli ATCC 25922. The n-hexane, aquadestilata, and ethyl acetate fractions of trembesi leaves with a concentration of 10% respectively had an average Inhibition zone diameters were 16.00 mm, 22.66 mm, and 3.23 mm against Escherichia coli ATCC 25922. The inhibition zone in the positive control was 21.80 mm, while the negative control used solvents for each fraction, namely n-hexane, aquadestylate, and ethyl acetate of 0.00 mm; 0.00 mm; 12.66 mm. The antibacterial activity was thought to be derived from the activity of flavonoid compounds, tannins, and triterpenoids contained in the trembesi leaf fraction.
