Abstract

Arabica coffee leaves are one of the traditional medicinal plants that have been trusted in healing various kinds of diseases. The pharmacological effects contained in coffee leaves have activity as an antibacterial agent, so it needs to be developed into a pharmaceutical preparation to increase ease of use, one of which is a cream preparation. This study aims to determine the arabica coffee leaf extract in cream preparations having antibacterial activity against Staphylococcus aureus in vitro using agar diffusion method. Staphylococcus aureus is a Gram positive bacterium in the form of cocci which is pathogenic and is involved in many skin infections. Every infected tissue, typical signs usually appear such as inflammation and abscess growth. The extraction of arabica coffee leaves was carried out using the soxhletation method with 70% ethanol. Arabica coffee leaf extract is made into cream preparations with concentrations of 10%, 20%, and 30%. Comparative antibiotic cream contained arabica coffee leaf extract is chloramphenicol cream. Chloramphenicol is an amphenicol antibiotic which is bactericidal with a broad-spectrum active activity against pathogenic bacteria. The cream preparations were tested for physical stability and antibacterial activity was known from the diameter of the inhibition zone around the disc. Data analysis was carried out by the Kolmogorov-Smirnov test, Levene statistic, One Way Anova and Spearman. The results of testing the antibacterial activity of arabica coffee leaf extract cream showed that arabica coffee leaf extract cream had antibacterial activity against Staphylococcus aureus. Cream of arabica coffee leaf extract with a concentration of 10%; 20%; and 30% respectively have an average inhibition zone of 9.25 mm; 14 mm; and 21 mm. Antibacterial activity is thought to originate from the activity of flavonoids, tannins and saponins in arabica coffee leaf extract. The arabica coffee leaf extract cream had a smaller inhibitory power compared to chloramphenicol cream which has an average inhibition zone of 37.75 mm. the results of One Way Anova analysis showed a significant difference between treatment groups (p <0.05). Arabica coffee leaf extract cream fulfills the organoleptic test requirements, homogeneity, pH, dispersion, adhesion, and protection power and is stable during the storage period.
