Abstract

In Indonesia many plants have the potential to be used as traditional medicinal ingredients, one of which is the cassava plant (Manihot esculenta, Crantz.). The leaves of cassava have potential as an antibacterial against Staphylococcus aureus which is a bacterium that causes infections in humans. One option for dealing with infections is to use antibacterial. Cassava leaf extract contains flavonoids, tannins, and saponins. Fractionation is needed to separate the compound based on its polarity. Based on the active compound content of cassava leaves, it is necessary to do research on the fraction of bacteria. The purpose of this study was to determine the antibacterial activity of cassava leaf fraction against Staphylococcus aureus and the concentration of fractions that had the greatest inhibition using the disk diffusion method. The research method used is experimental. Cassava leaf samples were extracted using maceration method with 70% ethanol. Then the results of maceration are concentrated and fractionation is done. The positive control used was chloramphenicol cream which is a broad-spectrum antibiotic and the negative control was DMSO. Cassava leaf fraction was made in series of concentrations of 1%, 3%, and 5%. The results of the antibacterial test were analyzed statistically by Kolmogorov-Smirnov, Levene Statistics, One Way Anova and Spearman. The results of antibacterial activity tests showed cassava leaf fraction has antibacterial activity against Staphylococcus aureus. Polar fraction of cassava leaves with a concentration of 1%; 3%; and 5%, respectively, have an average inhibition zone of 7 mm; 7.6 mm; and 8.83 mm. Semipolar fraction with a concentration of 1%; 3%; and 5%, respectively, have an average inhibition zone of 8 mm; 9 mm; and 10 mm. Whereas the nonpolar fraction has an average inhibition zone with a concentration of 1%; 3%; and 5% of 7.4 mm; 8.16 mm; and 8.6 mm. Antibacterial activity is thought to originate from the activity of flavonoid compounds, tannins and saponins in the fraction of cassava leaves. Cassava leaf fraction has a smaller inhibitory power than chloramphenicol cream which has an average inhibition zone of 30.6 mm. The concentration of fraction that has the largest inhibitory zone is a concentration of 5%. One Way Anova analysis results showed a significant difference between treatment groups (p<0,05).
