Abstract

Escherichia coli is a normal intestinal flora bacterial that is pathogenic in humans and can cause diarrhea if the number of bacteria in the digestive tract increases or is outside the body. Based on the pattern of causes of death of all ages, diarrhea was ranked 13th with a proportion of deaths of 3.5%. One of the efforts to prevent the disease is by using an antiseptic, but antiseptics on the market use chemicals as active substances so that in the long run it can irritate the skin. Therefore, it is necessary to have alternative preparations from plants that have higher benefits in terms of health. Papaya stems have the potential as an antibacterial against E. coli. The purpose of this study was to determine the antibacterial activity of papaya stem extract gel against E. coli using the agar diffusion method. The research method used is experimental. Papaya stem samples were extracted using the maceration method with 70% ethanol. Control the negative used is the gel base without extracts and positive control Hand sanitizer contains chloroxylenol. Papaya stem extract is made into gel preparations with concentrations of 5%, 10%, 15%, 20% and 25%. The gel preparations were subjected to physical evaluation which was organoleptic, pH, homogeneity, dispersal, adhesion, and protection for one month. Statistical analysis was performed with Shapiro-Wilk, levene statistics, One Way Anova and correlation. The results of the research conclusions showed that papaya stem extract gel with a concentration of 5%; 10%; 15%; 20%; 25% have antibacterial activity against Escherichia coli with the results of significant data analysis where the higher the concentration the wider the inhibitory zone. The antibacterial activity comes from the activity of flavonoid compounds, tannins, saponins, and anthraquinones in papaya stem extracts. Papaya stem extract gel has a smaller inhibitory power than the contains chloroxylenol gel which has an average inhibition zone of 31.6 ± 1.91 mm (very strong). Papaya stem extract gel meets organoleptic, homogeneous, pH, dispersion, adhesion, and protection test requirements and is stable in the storage period.
