Abstract

Bacteria Escherichia coli ATCC 25922 is a pathogenic microorganism that causes diarrheal disease, urinary tract infections, meningitis, and other infections. A common treatment for infectious diseases is antibiotics, but there is a lot of resistance to antibiotics caused by improper use. The case of resilience resulted in the need for alternative therapies for antibacterial agents derived from plants that have high potential as an antibacterial agent. Papaya stems have the potential as an antibacterial agent against Escherichia coli ATCC 25922 bacteria which are pathogenic bacteria that cause infections in humans. The purpose of this study was to determine the antibacterial activity of the papaya stem fraction against Escherichia coli ATCC 25922 by using the disc diffusion method and to find out the papaya stem fraction which has the highest inhibitory zone. The research method used is the experimental method. The research sample was papaya stems extracted using maceration method with 96% ethanol solvent. The extract was then fractionated using the liquid-liquid partition method using n-hexane, dichloromethane, and aquadestilate solvents. Papaya stem fraction is subjected to phytochemical screening to determine the compounds contained therein. The positive control used was erythromycin 500 mg tablet and the negative control used was each fraction solvent. Papaya stem fractions were each made with a concentration of 1%. Statistical analysis of research data was carried out using the Shapiro-Wilk, Kruskal-Wallis, and Mann-Whitney test methods. The results of the testing of the antibacterial activity of the papaya stem fraction showed the results that the papaya stem fraction had antibacterial activity against Escherichia coli ATCC 25922 bacteria. Papaya stem n-hexane, dichloromethane, and aquadestillate fractions with concentrations of 1% respectively have an average inhibition zone diameter of 12.57 ± 0.23 mm, 15.33 ± 0.35 mm, and 18.97 ± 0.58 mm against Escherichia coli ATCC 25922 bacteria. Antibacterial activity is thought to originate from the activity of flavonoid compounds, tannins, and saponins contained in papaya stem fractions.
