Abstract

Staphylococcus aureus is a positive Gram bacteria that has a natural habitat in humans, namely on the skin, the nasal mucosa, the mouth, and the colon. When the human immune system is weak, this bacteria can be pathogenic causing the conception, abscesses, and various pyrogen infections. Staphylococcus aureus is easily resistant to antibiotics and the amount of negative impact caused by the use of long-term antibiotics, then it takes a safer alternative to overcome the problem of Staphylococcus aureus infection namely by utilizing the active ingredient antimicrobial from medicinal plants. Water hyacinth is a plant that can incite bacterial activity. The purpose of this study was to determine the antibacterial activity of the extract maserate, the most active fraction of the ethanol fraction 96%, the dichloromethane fraction, the N-hexane fraction, and the optimum concentration of active water hyacinth fraction against Staphylococcus aureus. The leaf hyacinth is extracted using the maceration method with the 96% ethanol solvent and the process is fractionated using the solvent ethanol, dichloromethane, and N-hexane. Extracts and fractions of water hyacinth leaves are subjected to a phytochemical screening of flavonoids, alkaloids, and tannins. The antibacterial activity test is done by the diffusion method of paper discs with a concentration series of 15%, 30%, and 45%. Statistical analysis uses the one way ANOVA. Phytochemical screening results were positive for flavonoids, alkaloids, and tannins. Phytochemical screening of ethanol fraction and dichloromethane fraction was positive for the presence of flavonoid, alkaloid, and tannin compounds. The N-hexane fraction results in the negative presence of flavonoid compounds, alkaloids, and tannins. The results of the antibacterial activity test showed maserat of water hyacinth leaf extract has antibacterial activity with an average diameter of 10.3 mm. The ethanol and dichloromethane fraction has antibacterial activity with an average diameter of ethanol fraction of 21 mm and a dichloromethane fraction of 2.7 mm. Ethanol fraction is the most active fraction that has antibacterial activity against Staphylococcus aureus bacteria. The optimum concentration of the most active fraction is a 15% concentration with an average diameter of 13.67 mm. The concentration is the lowest concentration that can inhibit the growth of Staphylococcus aureus bacteria. Keywords: antibacterial, fractionation, leaf hyacinth, Staphylococcus aureus
