Abstract
Staphylococcus aureus ATCC 25923 is a bacterium that is easily found everywhere. It is known as pathogenic and often causes infection. In Jakarta in the period 1986- 1993, the incidence of Staphylococcus aureus ATCC 25923 infection increased nearly fourfold from 2.5% to 9.4%. Staphylococcus aureus ATCC 25923 causes various infections such as pneumonia, meningitis, empyema, endocarditis, acne, pyoderma, or impetigo. Antibiotics are one of the therapies used in dealing with infection, but the use of antibiotics has become a resistance due to improper use. Therefore, there is a need for alternative therapies from plants that have high potential as antibacterial. Papaya stems have potential as an antibacterial against Staphylococcus aureus ATCC 25293 which is an efection-causing bacteria in humans, to facilitate its use a medicine it is necessary formet it into pharmaceutical gel preparations. The purpose of this study was to determine the antibacterial activity of papaya stem extract gel against Staphylococcus aureus ATCC 25923 using the agar diffusion method. In this research, experimental method was used. Papaya stem samples were extracted using the maceration method with 70% ethanol. Clyndamicin gel used as a positive control and the negative control is the base gel without extract. Papaya stem extract was made into gel preparations with a concentration of 5%, 10%, and 20%. The gel preparations were evaluated including organoleptic test, pH, homogeneity, dispersion, adhesion, and protection for 10 days. Statistical analysis was carried out with data Normality test, Kruskal-Wallis, and Mann-Whitney. The results of the antibacterial activity of the papaya stem extract gel showed that the papaya stem extract gel had antibacterial activity against Staphylococcus aureus ATCC 25923. Papaya stem extract gel with a concentration of 5%; 10%; and 20% in sequence have an inhibition zone average of 19,7 ± 0.721 mm; 21,9 ± 1,625 mm; and 23,6 ± 1,629 mm. Antibacterial activity is thought to originate from the activity of flavonoid compounds, tannins, and saponins in papaya stem extract. Papaya stem extract gel has a smaller inhibition compared to Clindamycin gel which has an average inhibition zone of 28 ± 0,577 mm. Papaya stem extract gel fulfills organoleptic, homogeneous, pH, dispersion, adhesion, and protection and is stable during storage.
